PENGARUH DURASI LIGASI ARTERI CAROTIS COMMUNIS BILATERAL DENGAN REPERFUSI 1 JAM TERHADAP MORTALITAS TIKUS (Rattus novergicus)
Abstract
Latar Belakang : Stroke merupakan salahsatu sindrom penyakit penyebab disabilitas dan mortalitas nomer tiga di dunia. Prognosis stroke akan baik ketika masih berada dalam periode emas. Bilateral Common Carotid Artery Occlusion (BCCAO) dengan indikator mortalitas subjek, merupakan salahsatu tekhnik rat stroke model pada tingkat preklinik untuk meneliti periode emas dan pengembangan terapi stroke. Variasi teknik BCCAO dalam hal durasi ligasi/oklusi maupun reperfusi saat ini masih belum memiliki standar baku.
Tujuan Penelitian : Mengetahui pengaruh durasi ligasi arteri carotis communis bilateral dengan reperfusi 1 jam terhadap mortalitas tikus (Rattus norvegicus).
Metode Penelitian : Metode kuasi eksperimental posttest group design. Subjeknya menggunakan tikus jantan muda Rattus norvegicus galur wistar sebanyak 18 ekor, dibagi menjadi 3 kelompok. Kelompok-kelompok tersebut mendapat perlakuan durasi ligasi berturut-turut 5,10 dan 15 menit dengan durasi reperfusi 1 jam. Pencacatan mortalitas dilakukan di akhir penelitian. Analisis menggunakan uji chi square test dan dilanjutkan dengan uji fisher exact test dengan terlebih dahulu membagi tiga perbandingan analisis yaitu 5-15 menit, 5-10 menit dan 10-15 menit.
Hasil : Hasil uji chi square test menunjukkan hasil tidak memenuhi syarat, sehingga dilanjutkan dengan uji fisher exact test. Hasilnya adalah p>0,05 pada kelompok 5-10 menit dan 10-15 menit. Sementara untuk kelompok 5-15 menit tidak bisa dinilai karena menunjukkan hasil yang sama (tidak ada yang mengalami mortalitas).
Kesimpulan : Durasi ligasi arteri carotis communis bilateral dengan reperfusi 1 jam tidak berpengaruh terhadap mortalitas tikus.
Background : Stroke is one of the third leading disability and mortality syndromes in the world. The prognosis of stroke will be good when it is still in the golden period. The Bilateral Common Carotid Artery Occlusion (BCCAO) with subject mortality indicators, is one of the technique of rat stroke model at the preclinical level to examine the golden period and the development of stroke therapy. Variations of BCCAO techniques in terms of duration of ligation/occlusion and reperfusion are still not yet of standardized.
Research Objective : To determine the effect of bilateral carotid communis occlusion duration with 1 hour reperfusion on rat (Rattus norvegicus) mortality.
Research Methods : The quasi experimental method of posttest group design. The subjects used are 18 young male rats (Rattus norvegicus) wistar strain, divided into 3 groups. The groups were subjected to 5, 10 and 15 minutes duration ligation treatment with 1 hour reperfusion duration. Mortality was performed at the end of the study. The analysis using chi square test and continued with fisher exact test by firstly divided the three comparison of analysis that is 5-15 minutes, 5-10 minutes and 10-15 minutes.
Results : The result of chi square test shows that the result is not qualified, so it is continued with fisher exact test. The result is p> 0.05 in groups of 5-10 min and 10-15 min. While for the 5-15 minute group can not be assessed because it shows the same results (no mortality).
Conclusion : The bilateral carotid communis occlusion duration with 1 hour reperfusion did not affect rats (Rattus norvegicus) mortality.
