Preparasi dan Karakterisasi Nanosuspensi PLGA Kafein-Minyak Kemiri Menggunakan Solvent Evaporation Method
Abstract
Polymeric Nanoparticle using polymer PLGA (Poly Lactic-co-Glycolic Acid) is a widely developed drug delivery system in the last decade. PLGA is a copolymer of lactic acid (PLA) acid and poly glycolic acid (PGA) and can enhance drug penetration into the skin through the skin barrier. In this study, caffeine-kukui oil PLGA polymeric nanoparticle was formulated. The objective of this study was to investigate the preparation of PLGA nanoparticles as a carrier of caffeine-kukui oil and see the characteristics of PLGA nanoparticles as a carrier of caffeine-kukui oil. Solvent evaporation method was used in preparation. The method was done by mixing the organik phase into the water phase and was evaporated for 24 hours. The formulation uses D-Optimal optimization and in order to obtain a small particle size the amount of PLGA should be small and the amount of ethyl acetate and kukui oil should be greater. Characterization includes organoleptic observation, morphological observation, particle size measurement, polydispersity index, zeta potential value measurement, and % EE. Characterization of the resulted nanoparticles have particle size 413,5 ± 0,139 nm with spherical shape, polydispersity index of 0,503 ± 0,053, zeta potential value -47,33 ± 0,208 mv and efficiency percent yielded 61,04% ± 0,047. Solvent evaporation method can be used for the preparation of PLGA nanosuspension caffeine-kukui oil.
