FRAKSINASI DAN IDENTIFIKASI SENYAWA AKTIF PADA MENIRAN (Phyllantus niruri Linn) SERTA UJI AKTIVITAS SEBAGAI ANTIBAKTERI TERHADAP Pseudomonas aeruginosa
Abstract
Pneumonia is one type of grave infection caused by Pseudomonas aeruginosa bacteria. Pseudomonas aeruginosa resistance studies against several antibiotics have been found. So it is need to alternative treatment by using natural materials that are expected to be more effective, efficient, and safe, such as herbal meniran (Phyllantus niruri Linn). Meniran herbs contain bioactive components such as alkaloids, flavonoids, tannins, and saponins. This study was to ascertain the antibacterial activity against bacteria Pseudomonas aeruginosa. This study included extractions performed by extraction, evaporation, vacuum liquid chromatography, and thin layer chromatography (TLC). The result of each fractionation was antibacterial test and sub culture test. The fractions tested were ethyl acetate fraction: ethanol, n-hexane fraction: ethyl acetate and crude extract. The method used is the test of antibacterial activity with dilution. The test was performed by using multilevel series design with 2 concentrations of 10% and 5%, Amoxycilin positive control, negative paper disc control, DMSO 10% solvent control. Determination of KHM (minimum inhibitory concentration) is by testing using liquid microdilution method. The design of series of stratified with 4 treatments that is concentration 0.625%, 1.25%, 2.5%, 5%, negative control, paper disc, Amoxycilin positive control and 100% DMSO solvent control. The result of the microdilution was do by sub culture test and there was limpid zone at 5% concentration. The results of phytochemical tests were tested on alkaloids test, flavonoid test, saponin test, terpenoid test and phenol test. The results showed that the meniran extract from the ethyl acetate fraction: ethanol has antibacterial activity against Pseudomonas aeruginosa bacteria. The etil acetate fraction: 5% ethanol concentrations can inhibit Pseudomonas aeruginosa bacteria. This fraction shows that the most active fraction contains flavonoids. In a positive phytochemical screening test containing alkaloids, saponins, and total phenol.
