Uji Efektivitas Patch Transdermal Ekstrak Etil Asetat Batang Jatropha multifida L. terhadap Penyembuhan Luka Eksisi pada Tikus Jantan Galur Wistar
Abstract
Open wounds potentially cause infection that can lead to chronic infection. Jatropha multifida L. contains alkaloid, tannin, flavonoid, saponin and fenol that can accelerate the wound healing process. The purpose of study was to evaluate the effect of transdermal patch of ethyl acetate extract of J. multifida L. stem for excision wound healing. Wistar rats were used of experimental animals which divide into 6 groups, the control positive group treated with povidone-iodin 10%, the control negative group treated with base patch, group with 25 mg extract, and three other groups treated with patch of ethyl acetate extract of J. multifida L. with various concentrations (6,25 mg; 12,5 mg; 25 mg). Wounds were made based on Hongchang Gao method, which made 2 circles wound with diameter of ± 1 cm on the right and left of dorsal. Treatment and observation of wound healing were conducted during 14 days. The parameters observed were wounds width, the percentage of wound healing, the amount of fibroblast, neocapillary, and collagen density. The result showed that treatment of extract and patch of ethyl acetate extract of J. multifida L. with concentration 6,25 mg; 12,5 mg ad 25 mg decreased of wound width significantly on day 3,7,10 and 14 (p≤0,05). Percentage of wound healing data were analyzed usen the One Way ANNOVA, the result showed that treatment of extract and patch of ethyl acetate extract of J. multifida L. with concentration 6,25 mg; 12,5 mg and 25 mg has similar effect with povidone iodin 10% (p>0,05). The result of microscopic observation showed treatment with J. multifida could increase amount of neocapillary, fibroblast and collagen density were higger in patch 25 mg. It can be concluded that extract and patch of ethyl acetate extract of J. multifida L. can accerelate wound healing with the best activity in patch 25 mg.
