Abstract
Standardized red spinach leaf (Amaranthus triolor L.) in previous studies conducted on rat test animals has been shown to be an obesity preventive measure. At this time there has been no research on acute toxicity test on standard red spinach leaves. Therefore it is necessary to do this research to determine the LD50 value and the safety level of standard red spin leaf ethanol extract using the

Organization for Economic Cooperation and Development (OECD) method 425, and its effect on histopathology of kidney, liver and heart organ in male Sprague Dawley . In this study the mice were divided into the control group and the treatment group, the control group was given 2ml aquades and the treatment group was given standardized red spinach ethanolic extract with dose of 2000mg / KgBB. Mice observed individually include, signs of toxicity and body weight for 14 days. The results showed that there were no dead rats or signs of toxicity, so LD50 extract of red spinach ethanolic extract was more than 2000mg / KgBB and there was no significant change in rat weight (p≥0,05). The result of histopathology observation showed no change in the part of kidney, liver and heart organ from ethanolic extract of standard red spinach leaf dose 2000mg / KgBB, so that giving of ethanolic extract of standard red spinach leaves 2000mg / KgBB dose is considered safe.
